Overview

Rocakinogene sifuplasmid (INO-8000) is a therapeutic DNA plasmid vaccine developed by Inovio Pharmaceuticals in collaboration with the National Cancer Institute (NCI) for the treatment of chronic hepatitis C virus (HCV) infection. The vaccine consists of a consensus DNA sequence encoding the HCV non-structural proteins NS3, NS4A, NS4B, and NS5A from genotypes 1a and 1b. It is administered via intramuscular injection followed by in vivo electroporation, a delivery method that uses brief electrical pulses to create temporary pores in cell membranes, significantly enhancing the cellular uptake of the DNA plasmid and subsequent protein expression. This process aims to induce a robust, multi-antigen-specific cellular immune response, particularly CD8+ T cells, to target and clear HCV-infected cells. In clinical trials, rocakinogene sifuplasmid is often co-administered with INO-9012, a DNA plasmid encoding interleukin-12 (IL-12), which serves as an immunological adjuvant to further potentiate the T-cell response.
