Rocavorexant (formerly JNJ-61393215 or MAV-201) is a small molecule dual orexin receptor antagonist (DORA) being developed for the treatment of major depressive disorder (MDD). Developed by Janssen Pharmaceuticals in collaboration with Minerva Neurosciences, it targets both orexin-1 (OX1) and orexin-2 (OX2) receptors. Unlike traditional antidepressants, rocavorexant modulates the orexin system, which is involved in regulating wakefulness, arousal, and reward pathways. By blocking these receptors, it aims to improve symptoms of depression, particularly in patients with comorbid insomnia or those who have not responded to standard therapies.
