Overview

Rogletimide (pyridoglutethimide) is a second-generation nonsteroidal aromatase inhibitor that was investigated for the treatment of breast cancer. It is structurally related to glutethimide and aminoglutethimide but lacks significant sedative-hypnotic effects. Rogletimide functions by selectively inhibiting aromatase, the enzyme responsible for converting androgens to estrogens, thereby reducing estrogen levels in the body. Despite showing potential with fewer side effects than aminoglutethimide, rogletimide was never marketed due to its lower potency in clinical trials.
