ROI-001 is a preclinical-stage therapeutic candidate being developed by Revitope Oncology for the treatment of colorectal cancer (CRC). It is built upon Revitope's proprietary Precision Guided T-Cell Engager (P-TCE) platform, which utilizes a split-antibody approach to enhance tumor specificity. The technology involves two inactive components that are designed to bind to distinct antigens on the surface of a cancer cell. Only upon simultaneous binding to both targets do the components assemble into a functional T-cell engager that recruits and activates T-cells via the CD3 receptor to eliminate the tumor cell. This AND-gate logic is intended to reduce off-tumor toxicity by ensuring that T-cell activation only occurs in the presence of a specific dual-antigen signature unique to the cancer cells.
