ROI-004

ROI-004 is a preclinical-stage precision T-cell engager (TCE) being developed by Revitope Oncology for the treatment of solid and hematologic cancers. The therapeutic is built upon Revitope's proprietary TE-MOD (T-cell Engager Modular) platform, which utilizes a "split-antibody" architecture. This design involves two separate, inactive components that are engineered to bind to two distinct tumor-associated antigens. A functional T-cell engager is only formed when both components simultaneously bind to their respective targets on the surface of the same cancer cell. Once assembled, the molecule recruits and activates T-cells via the CD3 complex to induce targeted tumor cell lysis. This dual-antigen gating mechanism is intended to enhance tumor specificity and reduce off-tumor toxicity by sparing healthy tissues that do not co-express both target antigens.
