Drug intelligence / Profile preview
TQB2102 + TQB2450 (roldimatig deuderuxtecan + benmelstobart) is an investigational combination therapy being developed by Chia Tai Tianqing (a subsidiary of Sino Biopharmaceutical) for the treatment of HER2-expressing cancers, including HER2-positive advanced gastric or gastroesophageal junction (GEJ) cancer. TQB2102 (roldimatig deuderuxtecan) is a next-generation biparatopic antibody-drug conjugate (ADC) that simultaneously targets the extracellular domains 2 (ECD2) and 4 (ECD4) of HER2, conjugated to a deuterated topoisomerase I inhibitor payload via an enzyme-cleavable linker. TQB2450 (benmelstobart, also referred to as bemotrizumab) is a humanized IgG1 monoclonal antibody targeting programmed death-ligand 1 (PD-L1). By combining the targeted cytotoxic activity of the biparatopic HER2 ADC with the immune checkpoint inhibition of the anti-PD-L1 antibody, the combination aims to enhance antitumor efficacy, promote immunogenic cell death, and overcome resistance in HER2-expressing tumors.
