Overview

Rolinsatamab talirine (ABBV-176) is an antibody-drug conjugate (ADC) developed by AbbVie for the treatment of advanced solid tumors. It consists of a humanized monoclonal antibody (h16f/PR-1594804) targeting the prolactin receptor (PRLR), which is overexpressed in several solid tumor types. The antibody is conjugated to a highly potent DNA cross-linking pyrrolobenzodiazepine dimer cytotoxin (PBD; SGD-1882). The mechanism of action involves binding to PRLR on tumor cells and delivering the PBD payload intracellularly, resulting in DNA crosslinking and cell death. Preclinical studies showed activity against breast cancer models including triple-negative and low PRLR-expressing tumors. Clinical development included a phase 1 trial in patients with advanced solid tumors likely to express PRLR; however, this study was terminated due to significant toxicities such as cytopenias and cumulative effusions/edema[1][2][6].
