Overview

This therapeutic regimen is a novel multimodality immuno-chemotherapy platform being investigated by NYU Langone Health for the treatment of advanced mycosis fungoides and other cutaneous T-cell lymphomas (CTCL). The combination consists of the histone deacetylase (HDAC) inhibitor romidepsin, the Toll-like receptor 3 (TLR3) agonist poly ICLC (an immunostimulatory biologic), and focal lesional radiation. The regimen is designed to leverage the epigenetic effects of romidepsin to sensitize cancer cells, the immune-priming capabilities of poly ICLC to activate the innate immune system, and the potential abscopal effect of localized radiation to induce immunogenic cell death and systemic anti-tumor immune responses. It is primarily evaluated in patients with relapsed or refractory disease.
