Overview

Romosozumab is a humanized monoclonal antibody that targets and inhibits sclerostin, a regulatory protein involved in bone metabolism. By binding to sclerostin, romosozumab prevents it from inhibiting the Wnt signaling pathway in osteoblasts, thereby promoting bone formation and increasing bone mass. Additionally, inhibition of sclerostin reduces RANKL-mediated osteoclast activity, leading to decreased bone resorption. Romosozumab is primarily indicated for the treatment of osteoporosis in postmenopausal women at high risk for fracture—such as those with a history of osteoporotic fractures or multiple risk factors—or who have failed or are intolerant to other osteoporosis therapies. It is administered as a monthly subcutaneous injection and approved for use up to 12 months[1][2][3][5][6].
