RONDEL (RNAi/Oligonucleotide Nanoparticle Delivery) is a proprietary nanoparticle delivery platform developed by Calando Pharmaceuticals for the systemic administration of siRNA and other oligonucleotides. The system is built on a linear, cyclodextrin-containing polymer (CDP) that self-assembles with negatively charged siRNA to form nanoparticles typically less than 100 nm in diameter. These nanoparticles are designed to protect the RNA payload from nuclease degradation and prevent rapid renal clearance, while a polyethylene glycol (PEG) layer provides stability in the bloodstream. The platform often incorporates a targeting ligand, such as transferrin, to facilitate receptor-mediated endocytosis into specific cells, such as tumor cells. Once internalized, the siRNA is released into the cytoplasm to engage the RNA interference (RNAi) machinery. RONDEL was the first platform to enable the systemic delivery of siRNA in a human clinical trial via the candidate CALAA-01, which targeted the RRM2 protein in solid tumors.
