ropeginterferon alfa-2b + nivolumab is an investigational combination therapy being developed by PharmaEssentia for the treatment of hepatocellular carcinoma (HCC), particularly HBV-related HCC. The regimen consists of ropeginterferon alfa-2b (P1101), a novel, long-acting, mono-pegylated proline interferon, administered sequentially with nivolumab, an anti-PD-1 monoclonal antibody. Ropeginterferon alfa-2b acts as an agonist at the interferon alfa receptor (IFNAR), providing immune and antiproliferative effects, while nivolumab functions as an immune checkpoint inhibitor by blocking the PD-1 receptor. This combination is designed to enhance antitumor immunity and has demonstrated synergistic effects in suppressing HBV. It is currently being evaluated in Phase 1/2 clinical trials as an adjuvant treatment to prevent HCC recurrence following curative surgery.
