HYR-PB21 is a long-acting liposomal formulation of ropivacaine, an amide-type local anesthetic, developed for the management of postoperative pain. Ropivacaine functions by blocking voltage-gated sodium channels in nerve fibers, which inhibits the initiation and conduction of nerve impulses, thereby providing regional anesthesia and analgesia. By encapsulating ropivacaine within a liposomal delivery system, HYR-PB21 allows for a sustained release of the drug at the surgical site, extending the duration of pain relief compared to standard ropivacaine hydrochloride injections. This extended-release profile is intended to reduce the requirement for systemic analgesics, such as opioids, in the immediate postoperative period. The drug is primarily administered via local infiltration and has been evaluated in clinical trials for various surgical procedures, including inguinal hernia repair, hemorrhoidectomy, and bunionectomy.
