ROR1 bispecific refers to a class of bispecific T-cell engager (BiTE) antibodies designed to target the receptor tyrosine kinase-like orphan receptor 1 (ROR1) and the CD3 complex on T cells. ROR1 is an onco-embryonic antigen that is highly expressed in various hematological malignancies, such as chronic lymphocytic leukemia (CLL) and mantle cell lymphoma (MCL), as well as several solid tumors, including non-small cell lung cancer (NSCLC) and triple-negative breast cancer (TNBC), while remaining largely absent in healthy adult tissues. By simultaneously binding ROR1 on cancer cells and CD3 on T cells, these agents facilitate the formation of an immunological synapse, leading to T-cell activation and the subsequent lysis of the target tumor cells. The most clinically advanced candidate in this class is NVG-111, developed by NovalGen, which utilizes a humanized tandem single-chain variable fragment (scFv) format and targets a unique epitope on the Frizzled domain of ROR1.
