Overview

ROR1 CAR-T cells are an autologous cell therapy involving the genetic modification of a patient's own T lymphocytes to express a chimeric antigen receptor (CAR) specifically targeting the Receptor tyrosine kinase-like orphan receptor 1 (ROR1). ROR1 is a type 1 transmembrane tyrosine kinase receptor found on various malignancies, including triple-negative breast cancer, non-small cell lung cancer, chronic lymphocytic leukemia, mantle cell lymphoma, and acute lymphoblastic leukemia, while having limited expression on normal adult tissues. The CAR construct targets the Ig/Fz portion of ROR1's extracellular domain and incorporates 4-1BB and CD3ζ intracellular signaling domains to activate the T cells upon binding. The manufacturing process involves separating CD4 and CD8 T cell subsets from peripheral blood, culturing them, and transducing them with a lentiviral vector encoding the ROR1 CAR, resulting in a product formulated with a 1:1 ratio of CD4+ and CD8+ CAR-T cells. This therapy is being developed by Juno Therapeutics (a Bristol Myers Squibb company) and Fred Hutchinson Cancer Center.
