ROR1 CAR T cells with IL18 is an experimental chimeric antigen receptor (CAR) T-cell therapy developed by Miltenyi Biotec. It targets Receptor tyrosine kinase-like Orphan Receptor 1 (ROR1), a tumor-associated antigen expressed in various solid tumors, including pancreatic cancer. To overcome the immunosuppressive tumor microenvironment, these CAR T cells are armored with Interleukin-18 (IL18), either through constitutive expression or regulated by an NFAT promoter. IL18 enhancement is designed to improve CAR T cell expansion, persistence, and cytotoxicity. Preclinical studies in pancreatic cancer models have demonstrated that IL18-armored ROR1 CAR T cells exhibit superior anti-tumor efficacy and reduced exhaustion markers compared to standard ROR1 CAR T cells.
