Overview

ROR1-targeted genetically modified T cells are an investigational autologous chimeric antigen receptor (CAR) T-cell therapy developed by the University Hospital of Würzburg (Universitaetsklinikum Wuerzburg). These T cells are engineered to express a CAR that specifically targets Receptor Tyrosine Kinase Like Orphan Receptor 1 (ROR1), a type I orphan receptor protein. ROR1 is an oncofetal antigen that is highly expressed during embryonic development and in various malignancies, including chronic lymphocytic leukemia (CLL), mantle cell lymphoma (MCL), and solid tumors such as triple-negative breast cancer and ovarian cancer, while being largely absent in healthy adult tissues. The CAR construct typically utilizes the R12 scFv clone and a 4-1BB (CD137) costimulatory domain. Upon binding to ROR1 on the surface of cancer cells, the CAR T-cells undergo activation and expansion, leading to direct cytotoxic activity and tumor cell lysis.
