RORDEP (R. torques-derived endocrine peptides) polypeptides are microbiome-derived bioactive peptides synthesized by the commensal gut bacterium *Ruminococcus torques*. The family includes RORDEP1 (87 amino acids) and RORDEP2 (88 amino acids), which are found in human plasma and show an inverse correlation with adiposity. These peptides act as metabolic regulators that improve glucose tolerance, reduce weight gain, and increase bone density. Their mechanism of action involves the modulation of gut-derived hormones—specifically increasing plasma levels of GLP-1 and PYY while decreasing GIP—and the inhibition of hepatic glucose production via the gut-brain-liver axis. At the molecular level, RORDEPs downregulate gluconeogenesis, glycogenolysis, and lipogenesis in the liver while upregulating insulin signaling and glycogenesis. Developed by GutCRINE, these polypeptides are being investigated as pharmabiotics for the treatment of metabolic disorders including obesity and type 2 diabetes.
