Rosabulin (STA-5312) is a novel small molecule microtubule inhibitor with antimitotic and antineoplastic activity. It binds to tubulin at a unique site distinct from other agents such as vincristine, colchicine, or paclitaxel, inhibiting microtubule assembly and disrupting the cytoskeleton of tumor endothelial cells. This leads to cell cycle arrest and blockage of cell division, ultimately reducing blood flow to tumors and decreasing tumor proliferation. Rosabulin has demonstrated broad-spectrum anti-tumor activity in vitro against various solid tumors—including chemoresistant cancers—and in vivo in murine models. It was developed primarily for the treatment of advanced or metastatic solid tumors as well as hematological malignancies[1][2][3][4][5][7][9].
