ROSE12 + atezolizumab is an investigational combination therapy being developed by Chugai Pharmaceutical (a member of the Roche Group) for the treatment of locally advanced or metastatic solid tumors. The combination consists of ROSE12 (also known as RG6526 or RO7623066), a novel recycling monoclonal antibody targeting Glypican-3 (GPC3), and atezolizumab (Tecentriq), a humanized monoclonal antibody targeting programmed death-ligand 1 (PD-L1). ROSE12 utilizes Chugai's proprietary recycling antibody technology, which involves pH-dependent binding to allow the antibody to dissociate from its target in the endosome and return to the systemic circulation, potentially increasing its efficacy and duration of action. Atezolizumab functions as an immune checkpoint inhibitor, blocking the interaction between PD-L1 and its receptors (PD-1 and B7.1) to enhance the body's immune response against cancer cells. The combination is currently being evaluated in a Phase Ia/Ib clinical trial (NCT05907980) in patients with solid tumors that are refractory or resistant to standard therapies.
