Rosmantuzumab (OMP-131R10) is a humanized monoclonal antibody that targets R-spondin 3 (RSPO3), a protein involved in the modulation of the Wnt/β-catenin signaling pathway. By binding to RSPO3, rosmantuzumab inhibits its interaction with leucine-rich repeat-containing G-protein coupled receptors (LGR4, LGR5, and LGR6), thereby blocking activation of the RSPO-LGR pathway and curtailing aberrant Wnt/β-catenin signaling. This mechanism is implicated in various cancers and fibrotic diseases. Rosmantuzumab was developed by OncoMed Pharmaceuticals and has been investigated primarily for advanced solid tumors, including colorectal cancer[1][3][5][8].
