DW-5421 is a fixed-dose combination (FDC) small molecule therapeutic developed by **Daewon Pharm**, in collaboration with **Asan Medical Center**, for the treatment of hyperlipidemia. The formulation consists of **rosuvastatin**, an HMG-CoA reductase inhibitor (statin), and **ezetimibe**, a selective cholesterol absorption inhibitor. Rosuvastatin works by inhibiting the rate-limiting enzyme in hepatic cholesterol synthesis, which leads to an increase in LDL receptor expression and enhanced clearance of circulating LDL cholesterol. Ezetimibe complements this mechanism by targeting the **Niemann-Pick C1-like 1 (NPC1L1)** protein in the small intestine, thereby reducing the absorption of dietary and biliary cholesterol. This dual-action approach is designed to provide more robust lipid-lowering efficacy than statin monotherapy. DW-5421 is currently in Phase 3 clinical development in South Korea, potentially as a new strength or optimized formulation to expand the existing Crezet product line.
