Rosuvastatin + laroprovstat is a fixed-dose combination (FDC) therapy developed by AstraZeneca for the treatment of hypercholesterolemia. It combines rosuvastatin, a potent HMG-CoA reductase inhibitor (statin), with laroprovstat (also known as AZD0780), an oral small molecule inhibitor of Proprotein Convertase Subtilisin/Kexin Type 9 (PCSK9). While most PCSK9 inhibitors are monoclonal antibodies requiring subcutaneous injection, laroprovstat offers an oral alternative. The combination is designed to achieve profound reductions in LDL cholesterol (approximately 80% in Phase 1 studies) by simultaneously inhibiting cholesterol synthesis and increasing LDL receptor density on hepatocytes. It is currently in Phase 3 clinical development for patients with hypercholesterolemia, including those with heterozygous familial hypercholesterolemia.
