Drug intelligence / Profile preview
A locally-delivered, controlled-release formulation of rosuvastatin developed by the Government Dental College and Research Institute in Bangalore for the treatment of periodontal defects. The formulation consists of 1.2 mg of rosuvastatin (a synthetic, hydrophilic, second-generation statin) incorporated into a 1.2% methylcellulose-based in situ gel. Rosuvastatin acts as an HMG-CoA reductase inhibitor, which, when applied locally, exhibits pleiotropic effects including the stimulation of bone morphogenetic protein-2 (BMP-2) expression, promotion of osteoblast differentiation, and anti-inflammatory activity. In clinical trials (notably NCT02369250), this gel has been evaluated for the surgical treatment of mandibular Class II furcation defects, often in combination with autologous platelet-rich fibrin (PRF) and porous hydroxyapatite bone grafts to enhance clinical attachment level gain and bone fill.
