Overview

Rotigotine is an ergot-free dopamine agonist small molecule developed for continuous dopaminergic stimulation, most notably as a once-daily 24-hour transdermal patch marketed as Neupro. It acts primarily as an agonist at dopamine D3, D2, and D1 receptors and is used clinically for Parkinson's disease and moderate-to-severe primary restless legs syndrome; the patch formulation is designed to provide steady drug delivery over 24 hours. Rotigotine was originally developed by Aderis Pharmaceuticals and subsequently developed and commercialized by Schwarz Pharma, which became part of UCB. Beyond its approved neurology uses, clinical studies have also explored rotigotine in indications such as Alzheimer's disease, sleep and nocturnal motor symptoms associated with Parkinson's disease, and adolescent restless legs syndrome.
