Overview

Rottlerin is a natural polyphenolic compound isolated from the fruit of *Mallotus philippinensis* (the Kamala tree). It is widely utilized in biochemical research as a pharmacological tool. Historically, rottlerin was identified as a selective inhibitor of Protein Kinase C delta (PKCδ), although subsequent studies have demonstrated that it also acts as a mitochondrial uncoupler, which in turn activates AMPK and inhibits the PI3K/Akt/mTOR signaling pathway. In oncology research, rottlerin has been shown to induce autophagy and apoptosis in various cancer cell lines, including prostate cancer stem cells, by modulating these pathways. Beyond its research applications, rottlerin has traditional uses in Ayurvedic medicine for its anti-allergic and anti-helminthic properties.
