Overview

A combination therapy consisting of **rovalpituzumab tesirine** (an antibody-drug conjugate that targets DLL3 on tumor cells), **nivolumab** (an anti–PD-1 checkpoint inhibitor antibody), and **ipilimumab** (an anti–CTLA-4 checkpoint inhibitor antibody). Rovalpituzumab tesirine delivers cytotoxic agents directly to DLL3-expressing tumor cells, inducing cell death. Nivolumab blocks the PD-1 pathway on T cells, preventing antigen-mediated immune suppression and restoring antitumor T cell activity. Ipilimumab blocks CTLA-4, further enhancing T cell activation and proliferation. This combination is being explored mainly in the treatment of neuroendocrine tumors, including small cell lung cancer, aiming to potentiate immune response and deliver targeted cytotoxicity for improved antitumor effect.
