Overview

RP-72 (Rise Prot-72) is a small molecular weight recombinant protein that acts as an antagonist of the C-X-C chemokine receptor types 1 (CXCR1) and 2 (CXCR2). These receptors are the primary targets for interleukin 8 (IL-8, also known as CXCL8), a key mediator in inflammatory and tumor microenvironments. By binding directly to CXCR1/2, RP-72 inhibits the interaction between IL-8/CXCL8 and its receptors, thereby blocking downstream signal transduction pathways associated with inflammation, immune cell recruitment, and tumor cell proliferation. Preclinical studies have shown that this blockade can decrease proliferation of susceptible tumor cells—particularly pancreatic cancer cells—by interfering with CXCL8-mediated signaling. The drug is currently being investigated in early-phase clinical trials for its safety and efficacy as both monotherapy and in combination with gemcitabine for patients with advanced pancreatic cancer[1][3][5][6].
