Overview

**RP101 + gemcitabine** is an experimental combination therapy for cancer, most notably evaluated in advanced pancreatic cancer. **RP101** (also known as BVDU or (E)-5-(2-bromovinyl)-2'-deoxyuridine) is a small molecule nucleoside analogue that binds and modulates the activity of **heat shock protein Hsp27**, which is implicated in tumor cell resistance to chemotherapy and development of metastases. RP101 suppresses genes linked to drug resistance, assists in preventing chemoresistance, supports apoptosis, and enhances the efficacy of cytotoxic agents including gemcitabine. **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis and is a standard first-line chemotherapeutic for pancreatic and other cancers. In multiple studies, the co-administration of RP101 and gemcitabine increased remission rates, prolonged median survival, and delayed disease progression without major additional safety signals. The combination has advanced through phase 2 and phase 2b clinical studies for advanced pancreatic cancer.
