Overview

The rPA vaccine + alhydrogel is a combination anthrax subunit vaccine composed of recombinant Protective Antigen (rPA or rPA83), a protein component of *Bacillus anthracis* toxin, and the aluminum hydroxide-based adjuvant Alhydrogel. The recombinant PA is typically produced in *Escherichia coli* and purified for use. Alhydrogel acts as an immunologic adjuvant by forming a depot at the injection site and enhancing immune responses through increased antigen availability and activation of innate immunity pathways such as the NLRP3 inflammasome. The combination has demonstrated safety and immunogenicity in humans and provides protection against anthrax in animal models[1][2][5]. This formulation is under development primarily for prophylaxis against inhalational anthrax.
