Overview

RPH-051 is a monoclonal antibody developed by R-Pharm as a biosimilar to pertuzumab (Perjeta). Like its reference product, RPH-051 targets the Human Epidermal Growth Factor Receptor 2 (HER2), specifically binding to the extracellular dimerization domain (subdomain II). This mechanism prevents HER2 from forming heterodimers with other members of the HER family, most notably HER3, thereby inhibiting downstream signaling through the PI3K/Akt and MAPK pathways that drive cell proliferation and survival. It is primarily investigated for use in combination with trastuzumab and taxane-based chemotherapy for the first-line treatment of HER2-positive metastatic or locally recurrent unresectable breast cancer.
