RPH-120 is a fully human anti-PD-L1 IgG1 monoclonal antibody developed by R-Pharm for cancer immunotherapy. It is designed to target and block the interaction between Programmed death-ligand 1 (PD-L1) and its receptor PD-1, thereby preventing the suppression of T-cell-mediated anti-tumor immune responses. A distinguishing feature of RPH-120 is the inclusion of a specifically engineered Asn300Ala (N300A) mutation in its Fc fragment, which abolishes glycosylation and significantly reduces binding affinity to Fcγ receptors, thereby minimizing potential Fc-mediated effector functions such as antibody-dependent cellular cytotoxicity (ADCC). Preclinical studies have demonstrated that RPH-120 exhibits potent binding to human and non-human primate PD-L1 and achieves tumor growth inhibition comparable to approved PD-L1 inhibitors like atezolizumab. The drug is currently in clinical development for various oncology indications.
