RPI-78 is a catalytic RNA molecule, specifically a hammerhead ribozyme, designed to target and cleave the messenger RNA (mRNA) encoding Tumor Necrosis Factor-alpha (TNF-alpha). Developed by Ribozyme Pharmaceuticals, the drug was intended to treat inflammatory diseases, most notably rheumatoid arthritis, by preventing the synthesis of the TNF-alpha protein at the pre-translational level. This approach aimed to offer a more precise method of cytokine inhibition compared to traditional protein-based therapies like monoclonal antibodies. Clinical development reached Phase 1/2 trials in the late 1990s, but the program was ultimately discontinued as the company shifted its focus toward RNA interference (RNAi) technologies and was later acquired by Merck & Co.
