Drug intelligence / Profile preview
RPL19-25aa cryptic peptide is a novel circular RNA (circRNA)-derived peptide neoantigen encoded by circRPL19. It is highly expressed in immune checkpoint blockade (ICB)-responsive bladder cancer tissues and positively correlates with patient survival. RPL19-25aa acts by optimizing the electrostatic potential surface of the HLA-peptide-TCR complex, which increases complex stability and the TCR contact area, thereby enhancing HLA-TCR presentation. This optimization promotes dendritic cell-mediated CD8+ T-cell activation and induces interferon-gamma (IFN-γ) secretion, leading to enhanced antitumor immunity and tumor regression. It is being investigated as a potential target for personalized neoantigen-based cancer vaccines and immunotherapy combination strategies in bladder cancer.
