Overview

RPM CD19-mbIL15-CAR-T is a rapid personalized manufacture (RPM) chimeric antigen receptor T cell (CAR-T) therapy targeting the B-cell surface antigen CD19. The therapy uses autologous or donor-derived T cells that are genetically modified using the non-viral Sleeping Beauty transposon system to co-express a second-generation anti-CD19 CAR (comprising domains from CD8 and CD28), membrane-bound interleukin-15 (mbIL15), and a safety switch protein HER1t. The inclusion of mbIL15 is designed to enhance persistence and survival of memory stem-like T cells, potentially allowing for lower dosing and reduced risk of cytokine release syndrome. This approach enables manufacturing and infusion within two days after gene transfer, aiming to address relapsed/refractory B-cell malignancies in patients who have failed standard therapies including bone marrow transplantation[2][3][4][5].
