Overview

RPT1-CNS is a preclinical-stage small molecule hyperbolic inhibitor of nicotinamide phosphoribosyltransferase (NAMPT) developed by Remedy Plan Therapeutics. It is specifically designed for the treatment of glioma. Unlike traditional complete NAMPT inhibitors that cause severe on-target toxicities by shutting down NAD+ synthesis entirely, RPT1-CNS utilizes a unique hyperbolic inhibition mechanism. This approach partially decreases NAMPT activity to a level that selectively starves energy-demanding cancer cells of NAD+ while preserving sufficient metabolic function in healthy brain and central nervous system tissues.
