RPT1X is a preclinical-stage small molecule hyperbolic nicotinamide phosphoribosyltransferase (NAMPT) inhibitor being developed by Remedy Plan Therapeutics. NAMPT is a rate-limiting enzyme in the NAD+ salvage pathway, which is critical for cellular energy metabolism and is frequently dysregulated in cancer, autoimmune disorders, and metabolic diseases. Traditional NAMPT inhibitors completely shut down the enzyme, leading to severe on-target toxicities in healthy tissues. RPT1X utilizes Remedy Plan's proprietary hyperbolic inhibition mechanism to achieve fractional, tunable, and allosteric inhibition of NAMPT. This approach selectively reduces NAD+ levels below the survival threshold in diseased cells while preserving sufficient metabolic function in healthy cells, thereby avoiding systemic toxicity.
