RPTR-168 is an investigational autologous multi-targeted T cell (MTC) therapy developed by Repertoire Immune Medicines. It consists of genetically modified T lymphocytes derived from each patient’s own blood, which are primed and expanded against five tumor-associated antigens commonly expressed in HPV-16-positive tumors. These T cells are engineered to have interleukin-12 (IL-12) tethered to their surface using proprietary technology, enabling localized delivery of this potent immunostimulatory cytokine directly within the tumor microenvironment. The mechanism aims to enhance anti-tumor immune responses while minimizing systemic toxicity associated with IL-12. RPTR-168 is being evaluated in Phase 1 clinical trials for relapsed or refractory HPV-related carcinomas, melanoma, squamous cell carcinoma of the head and neck, and uterine cervical cancer[1][2][3][4][5].
