RQ-00201894 is a potent, selective, non-macrolide small molecule motilin receptor agonist being developed by RaQualia Pharma. It is designed to treat gastrointestinal motility disorders by stimulating gastric motility through the facilitation of cholinergic activity in stomach tissue. Unlike first-generation motilin agonists which often utilize complex macrolide structures (such as erythromycin), RQ-00201894 is a second-generation synthetic compound with an EC50 of 0.20 nM. It demonstrates high selectivity for the motilin receptor, showing over 90-fold selectivity compared to the ghrelin receptor. The drug has completed preclinical studies and is being evaluated for development in several indications, including gastroparesis, functional dyspepsia, and post-operative ileus.
