Overview

rQnestin34.5 V1 is an experimental oncolytic herpes simplex virus type 1 (oHSV-1) designed for the treatment of glioblastoma. It is engineered to express the viral ICP34.5 gene (a neurovirulence factor required for viral replication in non-dividing cells) under the control of a nestin promoter. Nestin is an intermediate filament protein that is highly expressed in glioblastoma and neural stem cells but absent in mature neurons, thereby restricting viral replication to tumor cells. This selective replication leads to direct oncolysis of the cancer cells and the subsequent release of tumor antigens, which can stimulate a systemic anti-tumor immune response. The "rQ" designation refers to the quiescent HSV-1 backbone used in its construction, which is designed to minimize neurotoxicity while maintaining potent oncolytic activity.
