Overview

RR-201 is a preclinical-stage bispecific antibody-drug conjugate (ADC) being developed by Roadrunner Therapeutics for the treatment of solid tumors, with a specific focus on refractory and drug-resistant cancers. As a bispecific ADC, RR-201 is designed to simultaneously bind to two distinct tumor-associated antigens, a strategy intended to enhance tumor specificity, increase internalization rates, and overcome resistance mechanisms common in traditional monospecific therapies. Upon binding to the target antigens on the surface of cancer cells, the conjugate is internalized, allowing for the intracellular release of a cytotoxic payload to induce cell death. Roadrunner Therapeutics is currently advancing RR-201 through preclinical studies and projects its first Investigational New Drug (IND) filing for its pipeline candidates in 2027.
