RROL antisense oligonucleotide is an experimental RNA-based therapeutic designed to target and deplete the long noncoding RNA (lncRNA) RROL (RNA Regulator of Lipogenesis). Developed by researchers at the Dana-Farber Cancer Institute, this antisense oligonucleotide (ASO) aims to treat multiple myeloma by disrupting a specific metabolic pathway. RROL functions as a molecular scaffold that facilitates the interaction between the MYC oncoprotein and the transcriptional modulator WDR82 at the promoter of the acetyl-CoA carboxylase 1 (ACC1) gene, a rate-limiting enzyme in de novo lipogenesis. By degrading RROL, the ASO prevents the formation of this transcriptional complex, thereby reducing ACC1 expression and inhibiting the lipid synthesis required for myeloma cell proliferation and survival.
