RRV RLI is an investigational viral gene therapy and immunotherapy designed for the treatment of glioblastoma (GBM) and other solid tumors. It utilizes a tumor-selective replicating retrovirus (RRV) vector to deliver a payload consisting of a superagonist interleukin-15 receptor-linked fusion protein (RLI). RLI is a fusion of IL-15 and the sushi domain of the IL-15 receptor alpha (IL-15Rα), which mimics the IL-15/IL-15Rα complex to potently stimulate immune effector cells. By leveraging the selective replication of the retrovirus in dividing tumor cells, the therapy transforms the tumor microenvironment into a local biofactory for RLI production. This localized expression promotes the infiltration and activation of CD8+ T cells and natural killer (NK) cells, overcoming the immunosuppressive nature of GBM. Preclinical studies have demonstrated significant survival benefits and synergistic effects when combined with standard-of-care chemotherapy like temozolomide.
