Overview

**RRx-001 + nivolumab** is an investigational combination therapy for advanced and refractory cancers. RRx-001 (bromonitrozidine) is a novel small molecule with pleiotropic mechanisms, notably functioning as a NLRP3 inflammasome inhibitor, an Nrf2 inducer, an epigenetic modulator, a nitric oxide donor, and a stimulator of tumor-associated macrophage repolarization. Nivolumab is a programmed death-1 (PD-1) immune checkpoint inhibitor monoclonal antibody. In pilot-phase clinical trials, this combination, administered intravenously, was shown to be safe and well-tolerated, with preliminary evidence of anti-cancer activity, including responses in tumors usually resistant to checkpoint inhibitors. RRx-001’s mechanisms include redox modulation, induction of reactive oxygen/nitrogen species, normalization of tumor vasculature, inhibition of CD47 and activation of epigenetic pathways, collectively aiming to boost and broaden the antitumor immune response when combined with nivolumab[3][5][1][2][4][6].
