RS-0461

RS-0461 is a preclinical-stage polymer-drug conjugate (PDC) developed by PDC Therapeutics (formerly RS Research) for the treatment of integrin-expressing solid tumors, with a primary focus on ovarian cancer. Constructed using the proprietary Sagitta® biodegradable nanocarrier platform, RS-0461 consists of the chemotherapeutic agent docetaxel conjugated to a polymer backbone that is functionalized with a ligand targeting integrin receptors (such as alpha-v beta-3). This targeted delivery approach is designed to enhance the therapeutic index of docetaxel by increasing its accumulation within the tumor microenvironment and facilitating receptor-mediated endocytosis, thereby reducing systemic toxicity and side effects like neutropenia. RS-0461 is part of a pipeline of targeted nanomedicines that includes RS-0139, a tumor-targeted docetaxel conjugate currently in clinical trials.
