RSV preF (specifically the version developed by Calder Biosciences, also known as DT-preF) is a protein subunit vaccine candidate designed to prevent Respiratory Syncytial Virus (RSV) infection. The vaccine utilizes Calder's proprietary 3D-Vaxlock technology, which employs dityrosine (DT) cross-linking to stabilize the RSV fusion (F) protein in its prefusion (preF) conformation. This stabilization is critical because the preF conformation is the primary target for potent neutralizing antibodies, yet it is naturally metastable and prone to irreversibly transitioning into a less protective postfusion state. By forming covalent, zero-length bonds between tyrosine residues, the DT cross-linking prevents this transition and enhances the protein's thermal stability. Preclinical data suggests that this stabilized antigen elicits a significantly more potent immune response compared to standard industry comparators. The program is currently in preclinical development, with planned indications for elderly populations and maternal vaccination to protect infants.
