Drug intelligence / Profile preview
RSV stabilized DT-preF (also known as DT-preF) is a preclinical-stage recombinant protein subunit vaccine being developed by Calder Biosciences for the prevention of respiratory syncytial virus (RSV) infections, particularly in older adults and maternal/infant populations. The vaccine utilizes Calder's proprietary 3D-Vaxlock dityrosine-crosslinking technology to introduce site-specific, covalent dityrosine bonds that lock the RSV fusion (F) glycoprotein in its highly immunogenic prefusion conformation (preF). This stabilization prevents the F protein from transitioning to its poorly protective postfusion state, thereby exposing key neutralizing epitopes (such as Site Ø and the Site IV/V interface). Preclinical studies have demonstrated that DT-preF elicits significantly higher neutralizing antibody titers compared to traditionally stabilized preF antigens (such as DS-Cav1) and can overcome immunosenescence in aged animal models when formulated with an alum adjuvant.
