RT-101

RT-101 is an oral biologic drug candidate developed by Rani Therapeutics that utilizes the proprietary RaniPill oral delivery platform to deliver octreotide, a synthetic peptide analog of somatostatin. The RaniPill is a sophisticated robotic capsule that, upon ingestion, protects the drug from gastric degradation and uses a micro-injection mechanism to deliver the payload directly into the intestinal wall, where it is absorbed into the systemic circulation. RT-101 functions as a somatostatin receptor agonist, mimicking the natural inhibitory effects of somatostatin on the secretion of growth hormone and other endocrine peptides. It was primarily investigated for the treatment of acromegaly and neuroendocrine tumors (NETs). While a Phase 1 study demonstrated that RT-101 achieved a relative bioavailability of approximately 65% compared to subcutaneous injection, Rani Therapeutics announced the discontinuation of the program in 2023 to prioritize other assets in its pipeline.
