RT-300 is a preclinical gene therapy candidate developed by Renova Therapeutics for the treatment of pulmonary hypertension and congestive heart failure. The therapy utilizes an adenoviral vector, specifically Ad5, to deliver the human urocortin 2 (UCN2) gene. Urocortin 2 is a potent peptide that selectively binds to and activates the corticotropin-releasing factor receptor 2 (CRF2 or CRHR2). Activation of this receptor in the cardiovascular system results in significant vasodilation and enhanced cardiac contractility (inotropy) and relaxation (lusitropy). While Renova's RT-200 program also employs UCN2 for heart failure and type 2 diabetes, RT-300 is specifically targeted toward pulmonary vascular indications, aiming to reduce pulmonary arterial pressure and improve right heart function.
