RT-4000 is an oral small-molecule dual agonist of the glucagon-like peptide-1 receptor (GLP-1R) and the gastric inhibitory polypeptide receptor (GIPR), currently under development by RenaissThera. Designed to address the limitations of injectable peptide-based therapies, RT-4000 aims to provide a cost-effective and scalable treatment for type 2 diabetes mellitus and obesity. The molecule was identified and optimized using an AI-powered platform to achieve potent dual agonism, which synergistically enhances insulin secretion and promotes weight loss. As of mid-2025, the program was in the lead optimization stage, with plans to advance toward IND-enabling studies.
