RTA 1701 is an orally bioavailable, small molecule allosteric inhibitor of nuclear receptor subfamily 1 group F member 3 (RORγt). It was originally developed by Reata Pharmaceuticals and is now under the ownership of Biogen following their acquisition of Reata. The drug acts as a selective antagonist of RORγt, a transcription factor critical for the differentiation of Th17 cells and production of interleukin-17A (IL-17A), which are implicated in autoimmune and inflammatory diseases. Preclinical studies demonstrated that RTA 1701 potently inhibits IL-17A expression in human immune cells and animal models, showing efficacy in models for rheumatoid arthritis and multiple sclerosis. Phase 1 clinical trials have been completed to evaluate its safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy volunteers[1][2][3][4][5].
